Dosage

FDA-labeled dosing: once-weekly subcutaneous injection, 5mg, 10mg, or 15mg, with a 20-week dose-escalation period before reaching maintenance dose in the obesity trial. This is the only compound in this batch with an actual FDA-labeled dose, not a community convention.

Side Effects

Most common: gastrointestinal (nausea, diarrhea, vomiting), predominantly mild to moderate, mainly during dose escalation. Discontinuation due to adverse events: 4.3% (5mg), 7.1% (10mg), 6.2% (15mg), vs. 2.6% placebo. Cardiovascular safety (SURPASS-4): MACE-4 hazard ratio 0.74 vs. insulin glargine — not elevated. Documented but rare: anaphylactic/allergic reactions.

Results Timeline

SURMOUNT-1 (obesity): 72-week trial with a 20-week dose-escalation period — most of the weight-loss curve happens after titration is complete. SURPASS-4 (diabetes): primary HbA1c endpoint at 52 weeks, safety follow-up to 104 weeks.
