LGD-4033 (Ligandrol) is a nonsteroidal, oral selective androgen receptor modulator (SARM) — one of the few compounds in this SARM category with an actual published Phase 1 human trial, often cited as the most-referenced SARM trial in the literature.

Human Phase 1 trial (n=76 healthy men, 21-50 years, 21 days, doses of 0.1/0.3/1.0mg daily): lean body mass increased dose-dependently (averaging 1.21kg at the 1.0mg dose) without a significant change in fat mass. Dosage

The human trial tested 0.1mg, 0.3mg, and 1.0mg daily, oral, for 21 days — the highest dose (1.0mg) produced the largest lean-mass gain but also the most pronounced hormone suppression (below).

Side Effects

Dose-dependent suppression of total testosterone and sex hormone-binding globulin was observed, with free testosterone and FSH significantly suppressed specifically at the 1.0mg dose. HDL cholesterol and triglycerides also decreased. Hormone levels and lipids returned to baseline after stopping treatment — the suppression wasn't permanent in this 21-day trial. No significant changes were found in hemoglobin, PSA, liver enzymes, or QT interval at any dose tested. Worth noting this trial only ran 21 days — it doesn't establish what happens with longer-term use.
